A sensitive bioassay for destruxins, cyclodepsipeptides from the culture filtrates of the entomopathogenic fungus Metarhizium anisopliae (Metsch.) Sorok. Anais da SEB

Destruxin A, a toxic cyclodepsipeptide, was purified from culture filtrates of the entomopathogenic fungus Metarhizium anisopliae (Metsch.) Sorok. by Reverse-Phase HPLC, and assayed for its effects on a larval Manduca sexta (L.) (Lepidoptera: Sphingidae) heart preparation. Destruxin A was found to cause reversible and dose-dependent acceleration of the heart beat. The heart bioassay was highly sensitive to pulse applications of destruxin A at concentrations as low as 35 pmol. Because of its high level of sensitivity, the M. sexta heart bioassay could be used to detect and quantify destruxins in tissue extracts from mycosed insects or to confirm bioactivity of HPLC fractions. Aortic body tumors in dogs from Northern Paraná, Brazil Ciência Rural

This article describes the occurrence of aortic body tumors in four dogs from Southern Brazil. All dogs were males, more than 9 year of age; most of these were from the Boxer breed of dogs. The animals had large neoplastic growths at the base of the heart, which demonstrated histologic characteristics consistent with that of aortic body tumor. By immunohistochemistry, tissue sections from all dogs reacted positively to neuron specific enolase. These cases might represent the first description of this tumor in dogs from Brazil, as confirmed by immunohistochemistry.

Cardiac and pulmonary alterations in symptomatic and asymptomatic dogs infectednaturally with Leishmania (Leishmania) chagasi BJMBR

Fifteen symptomatic and seven asymptomatic dogs infected naturally with Leishmania chagasi were examined in order to identify the presence of parasites and changes in heart and lung. Histopathological, cytological, and immunohistochemical analyses were performed on samples of heart and lung tissues. An inflammatory reaction characterized by inflammatory mononuclear, perivascular and intermuscular infiltrates was observed in both symptomatic and asymptomatic animals on histopathological analysis of the heart. Alkaline phosphatase (ALP) is important in calcification and its expression seems to be associated with the inflammatory process. We investigated the in vitro acute effects of compounds used for the prevention or treatment of cardiovascular diseases on total ALP activity from male Wistar rat heart homogenate. ALP activity was determined by quantifying, at 410 nm, the p-nitrophenol released from p-nitrophenylphosphate (substrate in Tris buffer, pH 10.4). Echocardiographic aspects of chronic mitral valvular disease were studied and compared to physical, radiographic and electrocardiographic aspects. Seventy dogs were used, and clinical examination, thoracic radiography, electrocardiogram and echocardiogram were performed. Correlations between regurgitation severity with cardiac failure functional class and murmur intensity were observed. The electrocardiogram showed a low sensibility in detecting cardiac chamber enlargement, caused by mitral regurgitation. All the dogs with severe mitral regurgitation showed cardiomegaly according to thoracic radiographies.

Acute nitric oxide synthase inhibition with N G-nitro-L-arginine methyl ester (L-NAME) on chronotropic and pressor responses was studied in anesthetized intact rats and rats submitted to partial and complete autonomic blockade. Abstract Lysosomal storage diseases (LSDs) are inherited conditions caused by impaired lysosomal function and consequent substrate storage, leading to a range of clinical manifestations, including cardiovascular disease. This may lead to significant symptoms and even cardiac failure, which is an important cause of death among patients. Currently available treatments do not completely correct cardiac involvement in the LSDs. Gene therapy has been tested as a therapeutic alternative with promising results for the heart disease. In this review, we present the results of different approaches of gene therapy for LSDs, mainly in animal models, and its effects in the heart, focusing on protocols with cardiac functional analysis.

Epilepsy-induced electrocardiographic alterations following cardiac ischemia and reperfusion in rats BJMBR

The present study evaluated electrocardiographic alterations in rats with epilepsy submitted to an acute myocardial infarction (AMI) model induced by cardiac ischemia and reperfusion. Rats were randomly divided into two groups: control (n=12) and epilepsy (n=14). It was found that rats with epilepsy presented a significant reduction in atrioventricular block incidence following the ischemia and reperfusion procedure. The aim of this study was to investigate the effects of training programs on serum lipid profile and myocardial oxidative stress. Male Wistar rats (2 mo-old) were divided into three groups (n=8): sedentary (S), loadless trained (T) and trained-overload 2% body weight (TL). T and TL were trained through swimming for 9 weeks. The objective of the present investigation was to determine the contractile effect of crude and acetone leaf extracts of Citrus sinensis (L.) Osb. on mammalian myocardium. Crude leaf extracts have been used in folk medicine to treat neurological disorders. Some flavonoids isolated from this plant presented a positive inotropic effect on myocardium. This motivated us to test the extracts on the atria of guinea pigs of both sexes (300-500 g) and surprisingly we observed inotropic depression instead of an increase in force.
